چکیده

Seeds collected at different maturation stages vary in physiological quality and patterns of protective antioxidant systems against deterioration. In this study we investigated the expression of genes that codify catalase (CAT), dismutase superoxide (SOD), and polyphenol oxidase (PPO) during the pre- and post-physiological maturation phases in whole seeds and in endosperms and embryos extracted from the seeds. Coffea arabica L. berries were collected at the green, yellowish-green, cherry, over-ripe, and dry stages, and the seeds were examined physiologically. The transcription levels of the genes were quantified by quantitative real-time polymerase chain reaction using coffee-specific primers. The highest level of SOD expression was observed in the endosperm at the cherry and over-ripe stages; in addition, these seeds presented the greatest physiological quality (assessed via germination test). The highest CAT3 transcript expression was observed at the green stage in whole seeds, and at the green and over-ripe stages in the embryos and endosperms. High expression of the PPO transcript was observed at the green and yellowish-green stages in whole seeds. In embryos and endosperms, peak expression of the PPO transcript was observed at the green stage; subsequently, peaks at the cherry and over-ripe stages were observed. We concluded that the expression patterns of the SOD and CAT3 transcripts were similar at the more advanced maturation stages, which corresponded to enhanced physiological seed quality. High expression of the PPO transcript at the over-ripe stage, also observed in the embryos and endosperms at the cherry stage, coincided with the highest physiological seed quality.

Effect of seed pre-treatment with L-arginine on improvement of seedling growth and alleviation of oxidative damage in canola plants subjected to salt stress. Fatemeh Nasibi1, 2*, Khosrow Manouchehri Kalantari1, 2 and Adeleh Barand1

Soil salinity is one of the major abiotic stresses that adversely affect plant productivity and quality. Therefore, an experiment was conducted to investigate the effects of seed treatment with L-arginine on some morphological and physiological parameters of Brassica napus under salinity stress. The seeds of canola were pre-treated with three arginine concentrations (0, 5, and 10µM Arg) for 24 ...

Gene expression of coffee seed oxidation and germination processes during drying.

Coffee (Coffea arabica L.) seeds are sensitive to desiccation and oxidative stress during drying processes. We investigated the effect of drying and moisture levels on germination-related gene expressions associated with enzymatic systems that prevent oxidative stress in coffee seeds. Coffee seeds collected at physiological maturity were subjected to slow and quick drying to 40, 30, 20, and 12%...

The effect of germination temperature on antioxidant enzymes activities and seed germination of Prangos ferulacea Seeds harvested at different seed moisture content

Prangos ferulacea is a plant from the umbeliferae family that is propagated by seed germination. In order to investigate the effects of seed ripening and germination temperature on germination and some antioxidant enzymes activity of this plant seed, a two-factor factorial experiment was carried out in a completely randomized design with three replications in the laboratory of Seed Sciences and...

Cucumber Response to Sphaerotheca fuliginea: Differences in Antioxidant Enzymes Activity and Pathogenesis-Related Gene Expression in Susceptible and Resistant Genotypes

Cucurbits powdery mildew is one of the most detrimental diseases of cucumber plants worldwide. A detailed insight into the biological processes leading to resistance or susceptibility to the pathogen would pave the road for an efficient disease-resistance breeding program. In the present study, the molecular and biochemical responses of a resistant vs. a susceptible cucumber cultivar infected w...

Changes in oligosaccharide content and antioxidant enzyme activities in developing bean seeds as related to acquisition of drying tolerance and seed quality.

Seeds of bean (Phaseolus vulgaris cv. Vernel) were collected throughout their development on the plant and dried at 15 degrees C and 75% relative humidity to a final moisture content of about 16% (fresh weight basis) to determine whether the onset of tolerance to this drying condition was related to changes in soluble sugars or the activities of the main antioxidant enzymes, namely superoxide d...
